Modern view on molecular bases of flaviviruses virulence from a position of their molecular-genetic properties investigated during last years by researchers of different countries was submitted. Known data of literature on influence of nucleotide and aminoacid replacements on neuroinvasiveness and neurovirulence of viruses are considered consistently in all proteins of flaviviruses genome. It was shown that both single, and multiple mutations in translated and not translated regions of flaviviruses genome could have the influence on virulence of a virus. The molecular characteristic in a complex with study of the basic biological properties of mutants in comparison with initial parental strain of a virus helps to understand the influence of separate replacements during the processes of variability of neurovirulence and neuroinvasiveness.
