Multilocus VNTR-genotyping of the cultures of Vibrio cholerae isolated in Kazan the outbreak of cholera in summer of 2001

Research Institute for Plague Control, Rostov-on-Don, Russia

The comparative study of the genomes of 80 cultures of V.cholerae eltor isolated in the focus of cholera in Kazan was made with the use of the VNTR-analysis and the single-primer polymerase chain reaction (PCR). According to the results of the analysis of the state of alleles in five VNTR-loci, 75 isolates were classified with one genotype, which was indicative of the clonal character of the outbreak. The profiles of polymorphic DNA fragments obtained in the single-primer PCR confirmed this conclusion. The shidy onthe possibility of using the multiplex variant of PCR for simultaneous analysis of microbial genomes in several loci of variable tandem repeats is in progress.
